Principle of the assay: human Chitinase 3-like 1 ELISA Kit was based on standard sandwich enzyme-linked immune-sorbent assay technology. A monoclonal antibody from mouse specific for Chitinase 3-like 1 has been precoated onto 96-well plates. Standards(NSO, Y22-T383) and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for Chitinase 3-like 1 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex was added and unbound conjugates were washed away with PBS or TBS buffer. HRP substrate TMB was used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the human Chitinase 3-like 1 amount of sample captured in plate.
Background: Chitinase-3-like protein 1, also called CHI3L1 or YKL-40 is a secreted glycoprotein that is approximately 40kDa in size that in humans is encoded by the CHI3L1 gene. This gene is mapped to 1q32.1. Chitinases catalyze the hydrolysis of chitin, which is an abundant glycopolymer found in insect exoskeletons and fungal cell walls. The glycoside hydrolase 18 family of chitinases includes eight human family members. This gene encodes a glycoprotein member of the glycosyl hydrolase 18 family. The protein lacks chitinase activity and is secreted by activated macrophages, chondrocytes, neutrophils and synovial cells. The protein is thought to play a role in the process of inflammation and tissue remodeling
